Can you remove them without opening anything?

No, and you should be wary of anyone who says they can. The comb, brood and honey are physically inside the cavity. A trap-out that lets bees leave but not return takes weeks, does not recover the queen reliably, and leaves every gram of wax and honey in your wall.

How much comb is actually in there?

It depends entirely on how long they have been there. A colony that moved in this spring might be a few small combs; one that has been in a roof for years can be many kilograms. That is the single biggest reason the price varies.
